Microfluidics are powerful tools for hemostasis and thrombosis research, enabling investigators to study cellular mechanisms and functional outcomes in controlled microenvironments that replicate key features of the vasculature. Precise control of geometry, shear flowFluid movement that applies force to cells and biofilms; used to mimic physiological/pathological conditions., cellular interactions, and biochemical conditions makes these systems valuable for basic research, drug development, and drug-safety assessment.
As demand for microfluidic assays grows, standardized methods are required to ensure consistent and comparable results across experiments and laboratories. To that end, the International Society of Thrombosis and Hemostasis (ISTH) Subcommittee on Biorheology, Biomaterials and Bioengineering published a list of recommendations and guidelines to standardize microfluidic assays, but there is a paucity of commercially available and readily accessible assays that conform to these guidelines. The purpose of this work was to develop and validate a standardized microfluidic clotting assay using a commercially available platform in accordance with ISTH recommendations.
This webinar will present the validation approach and demonstrate how the assay reproducibly evaluates platelet function, coagulation, and fibrinolysis under physiologically relevant flow conditions. Results will demonstrate that the validated protocol offers greater sensitivity and richer mechanistic and diagnostic insights than conventional methods, including platelet aggregometry, viscoelastometry, and thrombin- or fibrin-generation assays.
Attendees will learn how to:
- Incorporate a validated flow-based assay to their lab that was generated adhering to ISTH guidance criteria to improve assay consistency, comparability, and reproducibility.
- Standardize channel coating, apply adjustable shear-flow conditions, and perform multiparametric analysis.
- Set up, run, and analyze the assay using whole-blood and platelet-rich plasma samples for mechanistic research, therapeutic evaluation, drug-safety studies, and biomarker monitoring.
- Utilize the assay for better diagnostic capabilities and with greater sensitivity compared to other standard methods used in this field.
