Publication: Reenacting Neuroectodermal Exposure of Hematopoietic Progenitors Enables Scalable Production of Cryopreservable iPSC-Derived Human Microglia

In this study the CERO 3D Incubator & Bioreactor was used for large scale production of embryoid bodies form human iPSC’s, as well as to coculture cortical spheroid cultures containing iPSC derived microglia (iPSdMiG) for periods of 1 to 4 weeks. This study provides information on the production of human microglia, which are innate immune cells of the central nervous system, using induced pluripotent stem cells (iPSCs). The file presents an in vitro differentiation method for large-scale. production and cryopreservation of iPSC-derived microglia (iPSdMiG), which were shown to have a transcriptomic profile that demonstrated high similarity to adult human microglia, were immunopositivefor typical microglial markers, and were able to secrete pro-inflammatory cytokines, engage in phagocytotic activity, produce reactive oxygen species, and lend themselves to co-culture studies in neural 2D and 3D systems.
